                                                              western blot using anti-PIF3 antibodies

                                                              Materials: 4-days-old Col-0 and pifq Arabidopsis thaliana seedlings grown under dark (0.2 gram fresh weight) Methods: 0.2 gram of seedlings were extracted by the method described in Galvao et al. (2012). 20 µl of 600 µl plant extracts were separated on 8% Bis-Tris SDS-PAGE and blotted 1h to PVDF membrane. Blots were blocked with 5% milk in TBST for 1 h at room temperature (RT) with agitation. Blot was incubated in the primary antibody at a dilution of 1:1000 for overnight at 4 degree with agitation. The antibody solution was decanted and the blot was washed for 15 min with 3 times with TBST at RT with agitation. Blot was incubated in secondary antibody (anti-rabbit IgG HRP from BioRad) diluted to 1:5000 in for 1h at RT with agitation. The blot was washed as above and developed for 5 min with ThermoScientific PICO Chemiluminescent Substrate. Exposure time was 1 min.
