application example


western blot using anti-PIF4 antibodies
Total proteins were extracted from 0.2 gram fresh weight of Arabidopsis thaliana Col-0 and pifq seedlings grown under dark. 20 µl of 600 µl plant extracts were separated on 8% Bis-Tris SDS-PAGE using trank transfer 1 h to PVDF membrane. Blots were blocked with 5% milk in TBST for 1 h at room temperature (RT) with agitation. Blot was incubated with the anti-PIF4 antibody at a dilution of 1:1 000 for overnight at 4°C with agitation, washed for 5 min, 4 times with TBST at RT with agitation, incubated with the secondary antibody (anti-rabbit IgG-HRP) diluted to 1:5000 for 1 h at RT with agitation, and washed for 5 min, 4 times with TBST at RT with agitation. The blot was developed for 5 min with ThermoScientific DURA Chemiluminescent Substrate. Exposure time was 4 min.
