  Background:

Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) is an enzyme of ~37kDa that catalyzes the sixth step of glycolysis and thus serves to break down glucose for energy and carbon molecules. Plants contain both cytosolic and chloroplastic GAPDHs (glyceraldehyde-3-phosphate dehydrogenases). In Arabidopsis thaliana, cytosolic GAPDH is involved in the glycolytic pathway and is represented by two differentially expressed isoforms (GapC1 AT3G04120 and GapC2 AT1G13440) that are 98% identical in amino acid sequence.

Vulgare, Zea mays, Nicotiana tabacum, Populus trichocarpa, Gossypium raimondii, Cucumis sativus.

Lane 1: 5 μg total protein from Arabidopsis thaliana leaf. 
Lane 2: 10 μg total protein from Arabidopsis thaliana leaf. 
Lane 3: 25 μg total protein from Arabidopsis thaliana leaf. 
Electrophoresis: 15% SDS-Urea-PAGE
Transfer: blotting to NC (nitrocellulose) membrane for 1h.
Blocking: 5% skim milk at RT or 4℃ for 1h.
Primary antibody: 1:1000 dilution overnight at 4℃.
Secondary antibody: 1:20000 dilution using Goat Anti-Rabbit IgG H&L (HRP) (Cat# PHY6000)
Detection: using chemiluminescence substrate and image were captured with CCD camera.
