關(guān)蒼術(shù)HPLC指紋圖譜研究及白術(shù)內(nèi)酯Ⅰ和Ⅲ的含量分析

【摘要】 目的:建立關(guān)蒼術(shù)的高效液相色譜(HPLC)指紋圖譜,并測定關(guān)蒼術(shù)中白術(shù)內(nèi)酯Ⅰ和Ⅲ的含量。方法:指紋圖譜和含量測定研究的色譜柱分別為KromasilC18(250mm×4.6mm,5μm)和ODSHypersilC18(250mm×4.6mm,5μm),檢測波長分別為240nm和220nm,流動相分別為水-乙腈(梯度洗脫)和甲醇-水(60:40,V/V),流速為1mL.min-1,進(jìn)樣量為10μL。結(jié)果:關(guān)蒼術(shù)指紋圖譜中共標(biāo)定了9個共有指紋峰,10批樣品的相似度均>0.9。白術(shù)內(nèi)酯Ⅰ和Ⅲ的進(jìn)樣量分別在50.1～417.5ng(r=0.9997)和49.5～412.5ng(r=0.9998)范圍內(nèi)與各自峰面積積分值呈良好線性關(guān)系,平均加樣回收率分別為101.02%和97.12%,RSD分別為4.32%和4.01%(n均為9)。結(jié)論:所建指紋圖譜具有較好的精密度、重復(fù)性和穩(wěn)定性;所建含量測定方法簡單、靈敏、結(jié)果可靠,均可用于關(guān)蒼術(shù)的品質(zhì)評價和質(zhì)量控制。 更多還原

【Abstract】 OBJECTIVE:To establish HPLC fingerprint of Atractylodes japonica,and to determine the contents of atractylenolideⅠand atractylenolide Ⅲ in A.japonica.METHODS:Fingerprint study and content determination were performed on Kromasil C 18 column(250 mm×4.6 mm,5 μm)and ODS Hypersil C 18 column(250 mm×4.6 mm,5 μm);the detection wavelength was set at 240 nm and 220 nm;mobile phase consisted of water-acetonitrile(gradient elution)and methanol-water(60:40,V/V)at the flow rate of 1 mL.min-1.The injection volume was 10 μL.RESULTS:There were 9 common peaks in the fingerprints of A.japonica,and the similarity of 10 batches of samples was higher than 0.9.The linear ranges were 50.1～417.5 ng(r=0.999 7)for atractylenolideⅠand 49.5～412.5 ng(r=0.999 8)for atractylenolide Ⅲ.The average recoveries were 101.02% and 97.12%,and RSDs were 4.32% and 4.01%,respectively(n=9).CONCLUSION:Established fingerprint has good precision,reproducibility and stability;the method is simple,sensitive and reliable.It is suitable for quality evaluation and control of A.japonica.

【藥理作用】白術(shù)內(nèi)酯Ⅰ能促進(jìn)胃腸道消化吸收功能,是白術(shù)鍵脾的有效成分之-.具有抗炎、抗腫瘤作用，該成分還具有調(diào)節(jié)胃腸道功能和促進(jìn)營養(yǎng)物質(zhì)吸收的功能。
