用于檢測細(xì)胞凋亡的 Annexin V Kits
細(xì)胞膜磷脂不對(duì)稱性的喪失發(fā)生在細(xì)胞凋亡的早期。Annexin V 結(jié)合帶負(fù)電荷的磷脂如磷脂酰絲氨酸 (PS)。
Annexin V與暴露于細(xì)胞膜外層的PS的這種結(jié)合，已被作為檢測細(xì)胞凋亡的一種方法。Annexin V可以被熒光標(biāo)記，并常常與propidium iodide (PI) 一起經(jīng)由流式細(xì)胞儀測定以衡量細(xì)胞膜的完整性。
因此，沒有Annexin V 結(jié)合或 PI 染色的細(xì)胞代表健康的、有活力的細(xì)胞。呈現(xiàn)Annexin結(jié)合但沒有PI 染色的細(xì)胞可見于凋亡早期——細(xì)胞膜完整但有PS暴露。由于膜通透而結(jié)合Annexin V 和 PI 的細(xì)胞可見于凋亡晚期或細(xì)胞壞死。

Annexin V呈現(xiàn)抗磷脂酶活性并結(jié)合于磷脂酰絲氨酸。FITC標(biāo)記可通過流式細(xì)胞儀簡單直接檢測。Propidium iodide復(fù)染可用以區(qū)分凋亡細(xì)胞。 |
